Metabolite Identification
Common NameOrlistat
DescriptionOrlistat, also known as xenical or alli, belongs to the class of organic compounds known as leucine and derivatives. Leucine and derivatives are compounds containing leucine or a derivative thereof resulting from reaction of leucine at the amino group or the carboxy group, or from the replacement of any hydrogen of glycine by a heteroatom. A carboxylic ester resulting from the formal condensation of the carboxy group of N-formyl-L-leucine with the hydroxy group of (3S,4S)-3-hexyl-4-oxetan-2-one. Orlistat is a drug which is used for obesity management including weight loss and weight maintenance when used in conjunction with a reduced-calorie diet. also used to reduce the risk for weight regain after prior weight loss. use of orlistat is pending revision due to reports of liver-related adverse events. Orlistat is an extremely weak basic (essentially neutral) compound (based on its pKa).

General function:
Involved in monooxygenase activity
Specific function:
Cytochromes P450 are a group of heme-thiolate monooxygenases. In liver microsomes, this enzyme is involved in an NADPH-dependent electron transport pathway. It performs a variety of oxidation reactions (e.g. caffeine 8-oxidation, omeprazole sulphoxidation, midazolam 1'-hydroxylation and midazolam 4-hydroxylation) of structurally unrelated compounds, including steroids, fatty acids, and xenobiotics. Acts as a 1,8-cineole 2-exo-monooxygenase. The enzyme also hydroxylates etoposide.
Gene Name:
CYP3A4
