DescriptionThiotepa, also known as thioplex or tepadina, belongs to the class of organic compounds known as organic thiophosphoric acids and derivatives. These are organic compounds containing the thiophosphoric acid functional group or a derivative thereof, with the general structure RP(R')(R'')=S, where R,R',R'' = O,N, halogen residue. It is a stable derivative of N,N',N''- triethylenephosphoramide (TEPA). Thiotepa is an extremely weak basic (essentially neutral) compound (based on its pKa). Thiotepa is formally rated as a carcinogen (by IARC 1) and is also a potentially toxic compound. These drugs act nonspecifically. They stop tumor growth by crosslinking guanine nucleobases in DNA double-helix strands, directly attacking DNA.

Specific function:
Cytochromes P450 are a group of heme-thiolate monooxygenases. In liver microsomes, this enzyme is involved in an NADPH-dependent electron transport pathway. It performs a variety of oxidation reactions (e.g. caffeine 8-oxidation, omeprazole sulphoxidation, midazolam 1'-hydroxylation and midazolam 4-hydroxylation) of structurally unrelated compounds, including steroids, fatty acids, and xenobiotics. Acts as a 1,8-cineole 2-exo-monooxygenase. The enzyme also hydroxylates etoposide.
